Regulation of respiratory CD8+ T cell immunity by suppressive monocyte-like dendritic cells

چکیده
Abstract Active immune suppression can mediate the balance between protective cellular immunity and harmful immunopathology. This occur locally, at an infection site, or in regional draining lymph nodes (dLN). Immune regulation is of particular importance sites such as lung where aberrant immunopathology result loss tissue function. We have recently identified a novel population CD11b+CD103+CCR2+ monocyte-like dendritic cells (moDCs) which directly suppress CD8+ T cell proliferation vitro. Respiratory with RNA viruses recruits these moDCs either exclusively to dLN (after vesicular stomatitis virus infection) both site viral replication influenza infection). Exclusive depletion from using CCR2-DTR bone marrow chimeras results enhanced respiratory resident memory formation. By contrast, tract following responses coupled fatal Together, data suggest that suppressive govern key aspects by regulating function development long-lived cells, thereby balancing adverse pathology context infection. Simultaneous induction of CD4 T cell tolerance and CD8 T cell immunity by semimature dendritic cells.
Previous studies suggested that depending on their maturation state, dendritic cells (DC) could either induce T cell tolerance (immature and semimature DC) or T cell activation (mature DC). Pretreatment of C57BL/6 mice with encephalitogenic myelin oligodendrocyte glycoprotein (MOG)(35-55) peptide-loaded semimature DC protected from MOG-induced autoimmune encephalomyelitis. This protection was m...
متن کاملRegulation of immunity to respiratory syncytial virus by dendritic cells, toll-like receptors, and notch.
The activation and maintenance of pulmonary viral disease is regulated at multiple levels and determined by the early innate response to the pathogenic stimuli. Subsequent activation events that rely directly and indirectly on the virus itself can alter the development and severity of the ensuing immunopathologic responses. In the present review we outline several interconnected mechanisms that...

The identification of the specific DC subsets providing a critical role in presenting influenza antigens to naïve T cell precursors remains contentious and under considerable debate. Here we show that CD8(+) T lymphocyte (TCD8+) responses are severely hampered in C57BL/6 mice deficient in the transcription factor Batf3 after intranasal challenge with influenza A virus (IAV). This transcription ...
